Balancing Resolution, Field of View, and Depth of Field

Resolution, field of view, and depth of field are interdependent. A practical lens choice balances enough object-side resolution and coverage with usable depth of field for real inspection tolerances.

Balancing Resolution, Field of View, and Depth of Field

Resolution, field of view, and depth of field (DOF) are not independent checkboxes. In IVD imaging, the right lens balances object-side detail, coverage, focus tolerance, and throughput against the consumable and the mechanics.

Start with the target feature and the inspection task. If the target is a broad strip, cassette, plate, or microfluidic channel, a wide FOV can reduce tiling and increase UPH. If the target is a small cell or particle, resolution may dominate and a narrower FOV can be acceptable.

Resolution

Resolution must be high enough to separate the smallest clinically meaningful features, with margin for contrast, noise, illumination, and the sensor sampling limit. More magnification alone does not guarantee useful resolution: the complete optical chain and object-side pixel size matter.

Field of view

FOV determines how much sample is captured in one exposure. A larger FOV supports single-shot reading and simpler motion control, while a smaller FOV may be appropriate for high-detail inspection or tiled scanning. Match FOV to the active area and the location tolerances of the consumable.

Depth of field

DOF is the axial range that remains acceptably sharp. Real cartridges and plates can have warpage, meniscus variation, and assembly tolerances. A design with insufficient DOF may require precision Z motion and autofocus, increasing cycle time and BOM. Stopping down can improve DOF but costs light and may require more illumination.

Practical balance

Choose the lowest magnification that resolves the target with margin, then verify FOV, DOF, distortion, working distance, illumination, and sensor sampling together. Prototype with real customer consumables before tooling commitment.
